چکیده

Background Juvenile idiopathic arthritis (JIA) is a complex, multifactorial and chronic inflammatory disease of unknown etiology with considerable variability in which tumor necrosis factor-alpha (TNF-α) plays an important role. Both genetic and environmental factors can contribute to a susceptibility to disease initiation as well as a severity of disease course. TNF-α polymorphisms may be an independent marker of susceptibility and severity of JIA. The aim of this study was to elucidate putative association between the -863 C/A polymorphism in the promoter region of the TNF-a gene and susceptibility to onset and severity of JIA.

TNF-α Polymorphisms in Juvenile Idiopathic Arthritis: Which Potential Clinical Implications?

Whether tumor necrosis factor alpha (TNF-α) gene polymorphisms (SNPs) influence disease susceptibility and treatment of patients with juvenile idiopathic arthritis (JIA) is presently uncertain. TNF-α is one of the most important cytokine involved in JIA pathogenesis. Several single nucleotide polymorphisms (SNPs) have been identified within the region of the TNF-α gene but only a very small min...

Tumor necrosis factor alpha promoter polymorphisms in patients with juvenile idiopathic arthritis.

OBJECTIVE To investigate the potential association of tumor necrosis factor-alpha (TNF) promoter alleles within subtypes of juvenile idiopathic arthritis (JIA) compared to healthy controls in a Caucasian population. METHODS TNF-alpha promoter polymorphisms at positions -163, -238, -244, -308, -376 were determined in 228 patients with JIA and 196 healthy individuals. Genomic DNA was isolated a...

A Pilot Study of the Association of Tumor Necrosis Factor Alpha Polymorphisms with Psoriatic Arthritis in the Romanian Population

Tumor necrosis factor alpha (TNF-alpha) is an important pro-inflammatory cytokine implicated in the pathogenesis of psoriatic arthritis. We have performed a case-control association study of three TNF-alpha gene polymorphisms in a group of Romanian psoriatic arthritis patients versus ethnically matched controls. A second group of patients with undifferentiated spondyloarthritis was used in orde...

A tumour necrosis factor alpha polymorphism is not associated with rheumatoid arthritis.

OBJECTIVE To determine whether a polymorphism within the tumour necrosis factor alpha (TNF alpha) gene is associated with susceptibility to, or severity of, rheumatoid arthritis (RA). METHODS Consecutive patients with recent onset RA were enrolled in a prospective trial. DNA was collected, disease activity was measured at presentation, and radiographic progression at three years was assessed....

Guilt by association - what is the true risk of malignancy in children treated with etanercept for JIA?

Recently, the Food and Drug Administration placed a "black box" label on etanercept, and other tumor necrosis factor inhibitors used to treat childhood arthritis, warning of the risk of malignancies. The Food and Drug Administration made their decision based on a review of 48 cases of malignancies identified worldwide in children treated with tumor necrosis factor inhibitors for inflammatory bo...
